Historical Data-driven Formulation Design
Historical formulation development reports were reviewed to understand the established formulation system and guide the high-concentration formulation strategy.
Increasing the protein concentration of a bispecific antibody formulation can introduce additional formulation challenges, particularly when developing a high-concentration product intended for subcutaneous administration. Changes in concentration may affect formulation stability and require careful evaluation to ensure that product quality remains acceptable.
In this case, Protheragen supported the formulation development of a high-concentration bispecific antibody injection, with the protein concentration increased from 5 mg/mL to 150 mg/mL for development as a subcutaneous formulation.
The team reviewed historical process development reports and leveraged its experience in high-concentration formulation development to establish an appropriate formulation development strategy. Formulation confirmation demonstrated good stability of the high-concentration formulation. Furthermore, under elevated-temperature conditions, the degradation rates of the evaluated purity attributes showed no significant difference between the high- and low-concentration formulations.
Developing a Stable High-Concentration Subcutaneous Formulation
The key challenge was to increase the protein concentration from 5 mg/mL to 150 mg/mL while maintaining acceptable formulation stability.
The substantial increase in protein concentration could potentially affect the behavior and stability of the antibody formulation. Therefore, the formulation development strategy needed to identify a suitable formulation composition that could support the high-concentration product.
The development objectives were to:
The central development question was therefore:
Can the antibody concentration be substantially increased while maintaining a stability profile comparable to the established low-concentration formulation?
The team first reviewed the historical formulation development reports to understand the established formulation system and available development knowledge.
Historical development data provided a foundation for determining the appropriate formulation development direction and avoiding unnecessary experimental iterations.
This data-driven approach was particularly important because the project involved a substantial increase in protein concentration rather than a routine formulation adjustment.
The formulation strategy was developed by combining the historical project data with the team's experience in high-concentration biologics formulation development.
This experience was used to identify appropriate formulation considerations for the increased protein concentration and to establish a focused development strategy.
Rather than treating the 150 mg/mL formulation as a simple concentration adjustment, the development was approached as a high-concentration formulation optimization problem.
Based on the historical development information and high-concentration formulation experience, an appropriate formulation development strategy was established.
The selected formulation was subsequently subjected to confirmation studies to evaluate its stability and compare its degradation behavior with the original low-concentration formulation.
The formulation development successfully supported the increase in antibody concentration from: 5 mg/mL to 150 mg/mL for development as a high-concentration subcutaneous formulation.
This represents an approximately 30-fold increase in protein concentration.

Historical formulation development reports were reviewed to understand the established formulation system and guide the high-concentration formulation strategy.
The formulation strategy incorporated the team's experience in developing high-concentration biologics formulations to address the substantial increase in protein concentration.
The selected formulation was evaluated through long-term stability studies and high-temperature comparison studies to assess its stability and degradation behavior.
The antibody concentration was increased from 5 mg/mL to 150 mg/mL, supporting development of a high-concentration subcutaneous formulation.
The selected high-concentration formulation demonstrated good stability during the evaluated long-term stability study.
Under elevated-temperature conditions, the evaluated purity attributes showed no significant difference in degradation rates between the high- and low-concentration formulations.
The project successfully addressed the challenge of increasing the bispecific antibody concentration from 5 mg/mL to 150 mg/mL for development as a high-concentration subcutaneous formulation.
By combining historical formulation development data with high-concentration formulation expertise, Protheragen established an appropriate formulation development strategy and identified a suitable high-concentration formulation.
Formulation confirmation demonstrated good stability of the selected formulation. In addition, high-temperature stability comparison showed no significant difference in degradation rates of the evaluated purity attributes between the high- and low-concentration formulations.
The results supported the feasibility of the high-concentration formulation strategy and demonstrated the importance of stability-focused formulation development when substantially increasing antibody concentration.
