Glycosylation Profile Optimization

Glycosylation Profile Optimization

Glycosylation is an important critical quality attribute of therapeutic antibodies and can significantly influence product quality and comparability. In this project, the antibody product showed differences in glycan distribution compared with the reference product, particularly in G0F and galactosylated glycan levels.

Protheragen developed a customized upstream process optimization strategy combining CHO cell metabolism assessment, media formulation optimization, and cell culture process adjustment to improve the antibody glycosylation profile.

Challenge

The initial antibody product exhibited a glycosylation profile that differed from the reference product.

The main differences included:

  • Higher G0F content than the reference product
  • Lower galactosylation than the reference product
  • Insufficient consistency in the overall glycosylation profile

These differences affected the comparability of the antibody product with the reference product. The project therefore required targeted upstream process optimization to adjust the glycosylation profile while maintaining appropriate cell culture performance.

Our Approach

Protheragen leveraged its understanding of CHO cell metabolic pathways and antibody glycosylation to identify suitable opportunities for upstream process intervention.

The optimization program focused on two closely related areas:

This integrated approach helps reduce development risks, improve manufacturing consistency, and accelerate regulatory progression.

Customized Media Optimization

The culture medium composition was evaluated and optimized to provide a more suitable metabolic environment for the CHO cells and support the desired antibody glycosylation profile.

Cell Culture Process Optimization

Relevant cell culture conditions were subsequently adjusted in combination with the optimized medium. The process was evaluated based on both culture performance and the resulting antibody glycosylation profile.

This integrated approach enabled the upstream process to be tailored to the customer's specific glycosylation requirements.

Results

The customized production process successfully improved the antibody glycosylation profile. Compared with the initial product, the optimized process generated an antibody whose glycosylation distribution was highly consistent with the reference product - effectively closing the gap in both G0F and galactosylation levels.

Project Outcome

This case demonstrates the value of quality-attribute-driven upstream process development for controlling antibody glycosylation.

By integrating CHO cell metabolism knowledge, customized media development, and cell culture process optimization, Protheragen established a targeted production process that met the customer's specific glycosylation requirements and improved the similarity of the antibody product to the reference product.
