ADC Stability & Comparability Studies

ADC Stability & Comparability Studies

Antibody-drug conjugates (ADCs) are complex biotherapeutics whose quality can change during storage, transportation, formulation development, manufacturing changes, and process scale-up. Stability and comparability studies are therefore important for understanding product degradation, monitoring critical quality attributes, and evaluating whether changes introduced during development or manufacturing affect product quality.

Protheragen provides ADC stability and comparability studies using fit-for-purpose analytical strategies to monitor changes in molecular integrity, purity, aggregation, charge heterogeneity, DAR, free payload, biological activity, and other relevant quality attributes. Studies can be designed to support formulation development, process optimization, manufacturing changes, scale-up, technology transfer, and CMC development.

Overview of ADC Stability & Comparability Studies

ADC stability studies evaluate how product quality changes under defined storage and stress conditions, while comparability studies assess whether ADC materials produced under different conditions remain sufficiently similar in their relevant quality attributes.

Because ADCs contain an antibody, linker, and potent payload, degradation may occur through multiple pathways, including aggregation, fragmentation, deconjugation, payload-related degradation, oxidation, deamidation, and other molecular changes. A combination of orthogonal analytical methods is therefore used to monitor relevant quality attributes and identify meaningful changes over time.

Comparability assessments apply a similar analytical framework to compare materials generated before and after process changes, scale-up, manufacturing site transfer, formulation changes, or other development activities. The scope of testing can be tailored according to the nature of the change, development stage, and intended use of the study.

Our Services

Long-Term Stability Studies

Long-term studies monitor ADC quality under defined storage conditions over the intended study period. Testing focuses on changes in critical quality attributes and helps establish product stability profiles.

  • Protein concentration, purity, and molecular integrity
  • Aggregation, fragmentation, and charge variants
  • DAR, drug distribution, free payload, and biological activity

Accelerated & Stress Stability Studies

Accelerated and stress studies evaluate potential degradation under elevated or controlled conditions. These studies can support early formulation screening and identify quality attributes sensitive to environmental stress.

  • Temperature and other controlled stress conditions
  • Aggregation, degradation, and deconjugation monitoring
  • Identification of formulation or product vulnerabilities

Photostability & Temperature Stress Studies

Controlled light and temperature studies assess the susceptibility of ADCs to environmental factors that may affect molecular integrity or payload stability.

  • Light-induced degradation assessment
  • Temperature-related aggregation or fragmentation
  • Monitoring of DAR and payload-related changes

Forced Degradation Studies

Forced degradation intentionally subjects ADCs to selected chemical or physical stresses to generate and characterize degradation products. The resulting data can support degradation pathway investigation and stability-indicating method development.

  • Oxidative, thermal, pH, and light stress
  • Freeze-thaw and agitation studies, where applicable
  • Degradation pathway and product-related impurity assessment

Process & Manufacturing Comparability

Comparability studies evaluate ADC materials before and after manufacturing or process changes to determine whether relevant quality attributes remain consistent.

  • Conjugation and purification process changes
  • Process parameter and scale-up changes
  • DAR, purity, aggregation, charge variants, and other CQAs

Formulation & Process Change Comparability

Comparative studies assess the impact of formulation or process modifications on ADC quality. Analytical testing is tailored to the attributes potentially affected by the proposed change.

  • Buffer, excipient, pH, and concentration changes
  • Formulation optimization and process adjustments
  • Structural, physicochemical, and functional comparison

Manufacturing Site & Technology Transfer Comparability

Comparability assessments support manufacturing site changes and technology transfer by comparing ADC materials produced using different facilities or transferred processes.

  • Site-to-site analytical comparison
  • Pre- and post-transfer product assessment
  • Evaluation of process and product consistency

Stability-Indicating Characterization & Degradation Investigation

When changes are observed during stability or comparability studies, complementary characterization can be used to determine their molecular nature and potential impact on product quality.

  • Aggregation, fragmentation, and charge changes
  • Deconjugation and free-payload formation
  • Structural and functional investigation of observed differences

Key Quality Attributes for Comparability Assessment

The analytical scope of a comparability study is determined by the nature of the change and its potential impact on product quality.

Quality Attribute Representative Analytical Approaches Comparability Objective
Identity / Molecular Mass LC-MS / HRMS Confirm molecular consistency
Purity SEC-HPLC, CE-SDS, RP-HPLC Compare product-related species
Aggregation SEC-HPLC / UPLC Evaluate high-molecular-weight changes
Fragmentation CE-SDS, SEC, MS Assess molecular-size variants
Charge Variants CEX-HPLC, iCIEF Compare charge heterogeneity
DAR HIC-HPLC, LC-MS Compare drug loading
Drug Distribution HIC-HPLC, MS Evaluate conjugation heterogeneity
Conjugation Sites LC-MS/MS Compare conjugation patterns
Free Payload HPLC/UPLC, LC-MS Evaluate unconjugated payload
PTMs LC-MS/MS Compare molecular modifications
Higher-Order Structure Biophysical techniques Evaluate structural consistency
Biological Activity Binding / cell-based assays Compare functional properties

Not every study requires all attributes. The analytical panel is selected according to the type of change, development stage, risk assessment, and intended use of the comparison.

Stability & Comparability Study Workflow

Stability and comparability studies are designed according to the ADC molecule, development stage, study objectives, and potential impact of the conditions or changes being evaluated. A structured workflow integrates risk-based study design with orthogonal analytical testing to monitor relevant quality attributes, investigate observed changes, and support an overall assessment of product quality and consistency.
