Experience-driven Screening
Formulation development experience was leveraged to identify appropriate screening priorities and maximize the value of limited experimental opportunities.
Formulation development for bispecific antibodies can require careful screening and optimization because formulation conditions can influence protein stability and overall product quality. When the development timeline is highly constrained, an efficient screening strategy becomes particularly important, especially when only a limited number of screening rounds are available.
In this case, Protheragen supported formulation development for a bispecific antibody program under an accelerated timeline. Only two rounds of formulation screening were available, leaving limited tolerance for unsuccessful screening cycles.
By leveraging the team's formulation development experience and optimizing the screening workflow, a suitable formulation was identified in approximately 11 weeks, compared with a typical industry development timeframe of approximately 3–5 months. Subsequent formulation confirmation demonstrated good stability of the selected formulation, and the program successfully progressed to IND submission.
Limited Screening Opportunities Under an Aggressive Timeline
The primary challenge was the highly compressed development timeline.
Only two rounds of formulation screening were available for the project, resulting in a relatively low tolerance for unsuccessful screening cycles. A conventional screening workflow could increase the risk of consuming available development time before a suitable formulation was identified.
The formulation development strategy therefore needed to:
The key challenge was therefore not simply to screen more formulations, but to design a more efficient screening strategy within a very limited development window.
The formulation development team leveraged its previous experience with antibody formulation development to establish a focused screening strategy.
Rather than relying on an unnecessarily broad or sequential screening process, the team used existing formulation knowledge to determine appropriate screening priorities and streamline the experimental workflow.
This experience-based approach helped maximize the value of the two available screening rounds.
The screening workflow was rationally optimized to improve development efficiency.
The formulation candidates were evaluated through a structured screening process, allowing unsuitable candidates to be progressively eliminated and promising candidates to advance to subsequent evaluation.
The workflow was designed to make effective use of the limited experimental opportunities while maintaining sufficient evaluation of formulation stability.
By combining formulation development experience with an optimized screening workflow, the team was able to rapidly narrow the candidate formulations and identify a suitable formulation within approximately 11 weeks.
This accelerated approach helped reduce the development cycle while retaining a structured evaluation of formulation stability.
Suitable Formulation Identified in Approximately 11 Weeks
The optimized formulation screening workflow enabled the team to identify a suitable formulation in approximately: 11 Weeks
For comparison, the supplier's case information indicates that formulation development in the industry typically takes approximately: 3–5 Months
This represents a substantially shorter development cycle than the stated typical industry timeframe.
